The discovery of novel drug targets and the optimization of treatment regimens remain critical challenges in addressing diseases with unmet medical needs. Despite substantial progress in genetic and multi-omics research, existing methodologies often lack generalizability across diverse populations and fail to capture the complex, potentially non-linear relationships underlying drug responses. Specifically, most human genetics- facilitated drug target discovery efforts using Mendelian randomization (MR) rely on genetic resources from populations of predominantly European ancestry, potentially exacerbating health and healthcare disparities that affect underrepresented populations. Additionally, the dose-dependent and combinatorial effects of drug targets remain poorly understood, hampering the development of tailored treatments and precision medicine approaches. Several methodological challenges persist in achieving reliable drug target discovery and treatment optimization. For instance, confounding due to linkage disequilibrium (LD) undermines the validity of MR analyses, particularly in diverse populations with varying genetic architectures. Moreover, the absence of robust causal inference frameworks for populations with pre-existing health conditions complicates drug target discovery. In addition, biases in existing non-linear MR approaches hinder accurate evaluation of dose-response relationships, especially for therapeutic targets with saturation kinetics or other non-linear effects. To address these challenges, this program will develop and implement advanced statistical genetics tools over the next five years. From a theoretical perspective, we will focus on improving the robustness of MR to LD-induced confounding, ensuring more reliable causal inferences across diverse populations. We will design novel MR methodologies incorporating structural equation models for drug target discovery tailored to individuals with pre- existing health conditions, providing actionable insights for patient-specific therapies. Additionally, we will refine non-linear MR approaches to address known biases, thereby enabling more accurate and clinically relevant dose-response analyses. With these innovations, this program will also deliver transformative applications. We will conduct systematic drug target discovery by incorporating multi-omics and genetic resources from populations of diverse genetic ancestries, as well as populations with different combinations of pre-existing health conditions. We will elucidate the dose-dependent effects of the identified drug targets to optimize therapeutic dosing regimens for clinical application. Furthermore, we will extend the factorial MR framework to explore the effects of combined therapies, identifying potential synergistic interactions and supporting the development of precision treatment strategies. By integrating applied and theoretical advances, this program seeks to establish a robust, generalizable framework for identifying and optimizing drug targets. Ultimately, our goal is to bridge the gap between genetic discoveries and their clinical translation, advancing personalized medicine to improve health outcomes for diverse populations in the United States and worldwide.

Advancing Statistical Methods for Next-Generation Microbiome Data Analytics

TITLE: Advancing Statistical Methods for Next-Generation Microbiome Data Analytics Abstract: The focus of our research lab is to support human microbiome research with rigorous, robust, and practical statistical and quantitative methodology. In the next five years, our goals are to address critical data analysis gaps raised by recent advancements in microbiome epidemiology and bioinformatics, and to develop novel statistical methods that will facilitate the next generation of microbiome analytics. First, expanding microbiome consortia and public databases increase precision in testing the microbiome’s association with host conditions, but introduce more sources for unmeasured confounding such as population heterogeneity or uncollected covariates (e.g., medication). Recent statistical research has focused on confounding from the data’s compositional nature but largely ignored these unmeasured and potentially stronger factors. We propose to benchmark unmeasured confounding effects in microbiome studies with diverse real-world data, and to develop specialized latent factor modeling techniques for adjustment. This will improve false discovery control and facilitate robust findings in large-scale microbiome association studies. Second, modern bioinformatics can generate rich whole-microbiome genetic profiles with millions of microbial genes. But these profiles tend to have extreme levels of sparsity and lack functional annotations, limiting interpretability and statistical power in downstream findings. We propose to leverage recent breakthroughs in artificial intelligence and genomic large language models (LLMs) towards this problem. We will utilize the inherent functional dependency structure encoded by genomic LLMs to create putatively functional orthologs of microbial genes, which can meaningfully aggregate sparse and under-annotated “dark-matter” genes. This will empower downstream analyses and unlock the potential of modern microbiome bioinformatics. Third, metatranscriptomic (MTX) protocols are increasingly available, which characterize the microbiome beyond its functional capacities (“what can microbes do”) and reveal functional bioactivities in situ (“what are microbes doing”). MTX expressions depend on the underlying gene abundances, which are dynamic and error prone. Existing methods do not account for this and can generate biased findings. We will develop a rigorous error-in-variable model that will properly adjust for dynamic and noisy gene abundances, thus enabling robust differential expression analysis in new MTX studies. Our developed methodologies will be validated in different populations, disease settings, and microbiome ecologies, and implemented as open-source software. Derived data resources will be provided as publicly available databases. Through this proposal, our overall vision is to address critically unmet analytical needs from recent advancements in microbiome epidemiology and bioinformatics, and provide a publicly available toolkit of novel statistical methods that will support the next phase of human microbiome research and catalyze translational discoveries. 1

PROJECT SUMMARY Surgeon-scientists are uniquely positioned to bridge the gap between clinical care and research by translating surgical challenges into testable research questions and applying scientific discoveries to improve patient outcomes. Their unique position, combining surgical expertise, access to human biospecimens, and real-time exposure to disease processes, allows them to identify pressing clinical challenges and pursue solutions that are immediately relevant to human health. The contributions of surgeon-scientists have led to transformative innovations in fields such as organ transplantation, cochlear implantation, and cancer therapy. However, the surgeon-scientist workforce is rapidly declining due to limited access to structured training, increasing clinical demands, and a lack of protected time for research. This decline threatens continued innovation in Otolaryngology and communication sciences. The proposed program, Advancing Surgeon-Scientists through Career Education and Networked Training in Otolaryngology (ASCENT-Oto), aims to address this critical gap by providing structured research training, operative experience, and long-term mentorship to aspiring surgeon-scientists. ASCENT-Oto is designed for two target groups: post-residency Otolaryngology graduates who have not yet begun faculty careers, and early-career Otolaryngology faculty within two to five years of appointment. The program will offer a one-year mentored research experience for post-graduates and a 2.5-year K-equivalent training curriculum for early- career faculty. Participants will be matched to one of three sub-specialty research tracks aligned with NIDCD priorities: Hearing and Communication, Head and Neck Cancer, or Airway Health and Olfaction. Each track will integrate clinical and translational research with mentored surgical practice. Training will be supported by existing infrastructure at the Medical College of Wisconsin, the Clinical and Translational Science Institute, the MCW Graduate School, and OTO Clinomics, a department-wide clinical informatics platform. The program will feature individualized development plans, targeted didactic coursework, mentored research projects, and structured wellness programming to support professional growth and mitigate burnout. Each participant will be paired with one or more of twelve experienced mentors whose expertise spans basic and translational science, clinical trials, and outcomes research. Training will emphasize core research competencies, including hypothesis generation, study design, data analysis, scientific communication, and grant development. ASCENT-Oto is intended to build a pipeline of surgeon-scientists capable of sustaining dual careers in surgery and research. Program success will be evaluated through participant skill development, academic productivity, and long-term career outcomes. By preparing the next generation of clinician-investigators, ASCENT-Oto seeks to advance the mission of the NIDCD and improve the lives of individuals affected by disorders of hearing, voice, speech, and communication.

Advancing targeted delivery of intracellular protein therapeutics

PROJECT SUMMARY Intracellular protein delivery has immense potential for research and therapeutic applications, from replacing dysfunctional proteins to modulating key signaling pathways and safely delivering genome editing modalities. However, efficiently delivering functional proteins into the cell cytosol remains a significant challenge. Virus-like particles (VLPs), particularly those derived from retroviruses, offer a promising solution due to their ability to encapsulate large therapeutic proteins and directly deliver them into cells. These cell-derived nanoparticles, composed of assemblies of viral structural proteins such as “gag,” mimic natural viral delivery mechanisms while lacking viral genetic material. Researchers have begun leveraging this capability to transport diverse protein cargoes into cells. Despite their promise, the biogenesis and biological performance of VLPs are influenced by complex factors. For instance, packaging large macromolecules, distinct from the viral genomes they naturally encapsulate, can unpredictably alter their assembly properties. Similarly, their interactions with host factors and serum proteins during production impact assembly, stability, and cellular targeting. Moreover, limited understanding of how these factors influence VLP behavior in vivo, including the role of the protein corona, remains a critical barrier to their advancement as a viable delivery platform for research and clinical applications. To address these challenges, my laboratory will leverage our expertise in biomaterials design, nanoparticle- biological interactions, protein engineering, drug delivery, and nanomedicine to establish design principles for next-generation VLPs with enhanced targeting, potency, and bioproduction efficiency. First, we will develop heuristics for the optimal presentation of bioactive macromolecules on VLPs to enhance cell-specific uptake and targeting. Second, we will elucidate the composition of the VLP protein corona and its impact on in vivo targeting. By understanding how protein corona formation during production and systemic circulation influences VLP performance, we aim to refine their design for improved therapeutic outcomes. Third, we will identify genetic modifiers that regulate VLP assembly, cargo packaging, and release to optimize production yield, stability, and scalability for clinical applications. By elucidating the mechanisms underlying VLP assembly, functionalization, and tropism, the outcome of this work will be a set of design rules to advance the development of VLPs as versatile tools for research and therapeutic applications. These design rules will guide the creation of new VLPs capable of interfacing predictably with biological environments, paving the way for their clinical translation as targeted intracellular delivery platforms. Furthermore, our findings will contribute to the broader understanding and design of other cell-derived nanoparticles for drug delivery applications.

Since then, the Refuge Association and the Service have cooperated together to implement the Conserving the Future vision across the nation and across many different constituencies of the Refuge System.The National Wildlife Refuge Association is the only nonpartisan, nongovernment, independent organization exclusively focused on advancing the mission of the National Wildlife Refuge System. Building on a 41-year history of partnership, the Refuge Association works to amplify, assist and elevate the Refuge System s wildlife conservation and public engagement activities by providing expertise and assistance in partnership development, communications, outreach, education, land conservation and planning, as appropriate.In 2016 and 2017, we look forward to further collaborative work in our core areas of expertise and common interest: constituency-building, strategic habitat conservation and landscape conservation design, and communications and outreach. Advancing the Discipline of Translational Science: Professional Identity Development for Early-Stage Investigators

The Association for Clinical and Translational Science (ACTS) serves as the principal professional organization for the nation’s ~60 Clinical and Translational Science Award (CTSA) hubs. Each year, ACTS convenes over 1,200 faculty, trainees, and community stakeholders at its flagship Translational Science (TS) meeting, a premier forum for advancing the discipline of translational science. A central mission of ACTS is to elevate translational science as a rigorous, standalone discipline- a goal that aligns closely with the priorities of the National Center for Advancing Translational Sciences (NCATS), which has committed to sponsoring this application. As the field of translational science continues to evolve, a critical barrier to workforce development has emerged: the lack of professional identity development (PID) among early-stage investigators (ESIs). Evaluation data reveal that many ESIs gradually disengage from the field, often seeking mentorship and support within more traditionally defined disciplines. To address this gap, this R13/U13 application proposes a strategic initiative to embed PID into the professional development infrastructure of the TS meeting, thereby strengthening the translational science workforce. Our overarching goal is to cultivate a resilient, interdisciplinary cadre of translational scientists equipped to lead, collaborate, and innovate across boundaries. We propose two specific aims: Aim 1: Establish a professional development pathway for ESIs and mid-career investigators that addresses discipline-agnostic challenges and strengthens PID. We will design and implement a competency-aligned professional development pathway tailored to ESIs and mid-career investigators. This pathway will be open to all TS2026 attendees and will feature expert-led, skill- building workshops that address discipline-agnostic challenges. Each session will integrate PID principles through reflective exercises, peer-to-peer dialogue, and activities that align personal values with the ethos of translational science. Networking activities will include PID principles, providing opportunities for ESIs to explore their evolving roles, share experiences, and build supportive interdisciplinary partnerships spanning translational stages that reinforce a shared sense of purpose and belonging within the scientific community. Aim 2: Conduct a comprehensive, mixed-methods evaluation to assess impact and strengthen PID in CTS. We will launch a multi-year, mixed-methods evaluation framework to assess the impact of PID programming and inform continuous improvement. In Year 1 (the R13), we will conduct a qualitative study to define core PID characteristics and contextual factors unique to translational science. This evidence base will guide future programming and contribute to the broader understanding of PID in interdisciplinary research environments. By embedding PID into the professional development architecture of the TS meeting, ACTS will address a pressing national need: cultivating a robust, identity-driven translational science workforce capable of accelerating innovation and improving human health.

Advancing the Science of Social Connection Across the Life-Course: The Next Generation of Measurement in the United States

PROJECT ABSTRACT Social isolation, loneliness, and social connection are gaining global attention due to their profound impacts on the health and well-being of older adults. In 2023, advisories by the Office of the Surgeon General, legislation introduced by United States Senator Murphy and Representative Flood, and a Commission on Social Connection by the World Health Organization (WHO) all demonstrated urgency in addressing an “epidemic” of loneliness and social isolation. Central to advancing the field is updating existing measures of social connection and developing better instruments for assessing social isolation and loneliness that can fully capture the experience of today’s older adults. This two-day scientific meeting is therefore focused on advancing the science of measurement of social connection for older adults through an interdisciplinary approach. It will further support early-stage researchers and nurture national networking and collaboration. The following aims are proposed: Aim 1) To identify priorities in what should be included in measurements across settings, populations, and evaluation goals; Aim 2) To advance a national agenda to address gaps in measurement across aging populations, creating a foundation for future discussions and conferences; and Aim 3) To provide mentorship to, and create community amongst, early-stage investigators focused on social connection research. The scientific meeting will bring together US-based researchers, decision-makers, and practitioners with expertise in social connection across varied scientific disciplines (e.g. social and health sciences) and interested parties (e.g. academia, community-based, policy, industry). Our approach will largely rely on a workshop format to enable open discussion on the major challenges and gaps in measurement across settings, priorities for aligning measures, and innovations in methods to gather data. We will support early-stage investigators through travel stipends, mentor-mentee pairing, and an aims workshop. In addition, we will generate clear recommendations for multiple interested parties through a peer-reviewed convening paper, a policy brief, and online materials. Taken together, the scientific meeting will serve as a critical starting point to advance the science of loneliness, social isolation, and social connection measurement among older adults. It will build meaningful national collaborations, support early-stage investigators, and inform measurement considerations for funders of social connection research. This conference will serve as a basis for future R13 applications to hold interdisciplinary national meetings, including around innovative interventions and technologies.

Advancing Transplantation Outcomes in Children

Project Summary/Abstract Renal transplantation is widely recognized as the treatment of choice for children with end stage renal disease (ESRD). The life expectancy benefit is significant and a functioning renal transplant enables children to grow well, develop almost normally and improve their school educational performance levels. However, current data indicate that virtually all grafts in pediatric recipients will eventually fail due to chronic allograft dysfunction, and as such, the goal of preserving long-term allograft function is the key area for future progress. Furthermore, registry data indicate that opportunistic infections are now the most common cause for hospitalization and death in the pediatric population. Little is known about pathogen-specific protective immunity in pediatric recipients who are exposed to multiple novel infectious agents throughout the post-transplant period in the absence of Tmemory. Our approach in this trial is based on the concept that successful preservation of long-term allograft function requires an immunosuppressive regimen that targets donor specific alloantibody (DSA) production while preserving pathogen-specific immunity. We also propose that pediatric recipients require precision tools to monitor, identify and prevent silent subclinical intragraft inflammation/rejection, which is common at early times in the post transplant period. Based on a recent pilot study using de novo Belatacept therapy in combination with an mTOR inhibitor (mTORi) in pediatric recipients, we will test the hypothesis that early introduction of a Belatacept/sirolimus maintenance immunosuppressive regimen is safe and efficacious in children to augment immunoregulation, prevent DSA production and enhance long-term allograft function. EBV seropositive primary renal transplant recipients, aged between 6 and 21 yrs, from eleven experienced pediatric clinical centers will be randomized to receive induction therapy with anti-thymocyte globulin and either Belatacept therapy in combination with sirolimus or remain on standard immunosuppression therapy using tacrolimus and mycophenolate mofetil. Primary endpoint analysis includes de novo DSA development and assessment of allograft function after 36 months of follow up. Associated studies include surveillance monitoring using a novel automated point-of-care urine biomarker assay, and in-depth mechanistic studies on the cellular basis for pathogen-specific immunity and evaluation of functional antibody responses to vaccine. Extensive mechanistic studies will also be performed to assess the impact of Belatacept/mTORi on cellular and humoral alloimmunity and the further development of urinary biomarkers to differentiate subclinical rejection from infection. There are significant unmet clinical needs in pediatric recipients who have unique pathogen-specific and alloimmune responses following transplantation. Overall, the relevance of this proposal is that it builds upon previous trials to test if a novel agent (Belatacept) targets allograft dysfunction; in-depth mechanistic monitoring will allow for the prediction of patient course, and our findings will be applicable to recipients of other solid organ transplants.

Aeronomy

The Aeronomy Program supports research from the mesosphere to the outer reaches of the thermosphere and all regions of the Earth s ionosphere. The Aeronomy Program seeks to understand phenomena of ionization, recombination, chemical reaction, photo emission, and the transport of energy, and momentum within and between these regions.The program also supports research into the coupling of this global system to the stratosphere below and magnetosphere above and the plasma physics of phenomena manifested in the coupled ionosphere-magnetosphere system, including the effects of high-power radio wave modification. The Coupling, Energetics, and Dynamics of Atmospheric Regions (CEDAR) Program aims to understand changes in the atmosphere over short and long time scales. CEDAR is consistent with the recommendations and goals of the NAS Decadal Survey "Solar and Space Physics: A Science for a Technological Society". A primary goal of CEDAR is to explain how energy is transferred between atmospheric regions by combining a comprehensive observational program with theoretical and empirical modeling efforts. A data base of CEDAR observations is maintained for community use. The annual CEDAR Workshop attracts over 300 scientists including a large number of graduate students and as well as many international collaborators.
