Abstract Significance: HIV persists in long-lived CD4⁺ T cell reservoirs despite suppressive ART, as integrated proviruses remain poised for reactivation. Chromatin remodeling is a central barrier to durable silencing, yet most studies have focused on SWI/SNF family members. The roles of non- SWI/SNF remodelers remain poorly defined, limiting our ability to rationally design host-directed “block-and-lock” cure strategies. Our unbiased shRNA screen of all 16 human remodeler ATPases identified EP400, CHD1, and CHD9 as repressors and INO80A, SMARCA5, and CHD2 as activators, establishing chromatin remodeling as a key determinant of HIV latency. Innovation: Our prior studies revealed that the p400 complex regulates HIV transcription through dual mechanisms: directly, by engaging Tat via the DMAP1 subunit to block Tat-TAR RNA interactions and restrict p-TEFb recruitment; and indirectly, by altering host transcriptional programs that control T cell activation states. Building on this mechanistic precedent and methodological platform, we now focus on INO80A, SMARCA5, CHD1, and CHD2, remodelers from distinct ATPase families that govern Tat-independent checkpoints at initiation, pause release, and elongation. Methodologically, we will apply TurboID-ChAP-MS (locus-specific proteomics), BEM-seq (single-nucleosome mapping), and degron-mediated acute depletion with ATPase-dead rescue to interrogate remodeler function with unprecedented resolution. Approach: Aim 1 will define the ATPase requirement and transcriptional checkpoints regulated by INO80A, SMARCA5, CHD1, and CHD2 using degron/CRISPR perturbations, ChIP-seq, nascent RNA profiling, and nucleosome mapping. Aim 2 will characterize remodeler-specific complexes and Tat dependence at the HIV promoter via TurboID proximity labeling integrated with chromatin affinity purification-mass spectrometry. Aim 3 will test combinatorial perturbations in Jurkat and primary CD4⁺ T cell latency models, including ART-suppressed donor cells, to identify synergistic “block-and-lock” strategies that enforce durable proviral silencing. Impact: By defining remodeler-specific mechanisms at discrete transcriptional checkpoints and leveraging their enzymatic, druggable activities, this work will establish chromatin remodeling as a therapeutic axis for durable HIV suppression and functional cure.

ABSTRACT Significance to VA: Knee osteoarthritis is a major health problem in Veterans and accounts for approximately one-half of all joint replacement surgeries in the VA system. Total knee arthroplasty is the current standard of care to treat advanced knee osteoarthritis and is effective in reducing pain. However, nearly 1 in 4 patients report not being satisfied with their surgery. While the reasons for this high number of unsatisfied patients is multifactorial, poor knee function is a major driver behind poor outcomes. Clinical research studies often characterize functional deficits by evaluating Arthrogenic Muscle Inhibition (AMI). But these evaluations typically require the patient to maximally contract their muscles, which often causes pain and leads to unreliable results. Additionally, specialized equipment and highly trained investigators are essential for reliable measurements. Innovation and Impact: This project addresses these challenges in quantifying AMI by developing, validating, and implementing a novel nerve stimulating platform called the Smart Nerve Stimulator to quantify Arthrogenic Muscle Inhibition (AMI) in Veterans before and after undergoing total knee arthroplasty surgery. This is an impactful innovation because it addresses an unmet clinical need by leveraging state-of-the-art electrode technology and a custom-developed user interface to establish the first-of-its-kind neuromechanical database in Veterans with knee pain. We expect that this will serve as a novel biomarker of knee health and support future VA research aimed at improving therapeutics for Veterans with knee osteoarthritis. Specific Aims: This device development Merit application has 3 aims. Aim 1 will optimize a fully automated nerve conduction testing tool using custom developed software and high-density stimulating and recording electrode arrays. Aim 2 will establish the first-of-its-kind Neuromechanical Database to Advance Veteran Health. Aim 3 will develop a novel AMI biomarker using machine learning. Methodology: This study uses an iterative design cycle in Aim 1 to optimize a working prototype that leverages our automated software interface and designs, fabricates, and deploys high-density stimulating and recording electrodes. Our Smart Nerve Stimulator will quantify AMI as the ratio H-reflex:M-max that represents the spinal and muscle excitability. Aim 2 uses a cross-sectional study design to evaluate arthrogenic muscle inhibition in 3 Veteran groups: 1) Veterans with knee osteoarthritis who are scheduled to undergo total knee arthroplasty surgery, 2) Veterans who are age/sex-matched but without any knee symptoms or pathology, and 3) Veterans who are 25-45 years of age and sex-matched without any knee symptoms or pathology. Aim 3 uses our innovative biomechanically informed latent neuromechanical dynamics learning architecture to automatically differentiate between Veterans with knee osteoarthritis, total knee arthroplasty, and healthy controls. We will quantify arthrogenic muscle inhibition before and after total knee arthroplasty to evaluate the effects of surgery on neuromechanical function. We will also quantify arthrogenic muscle inhibition with young and age-matched Veterans without knee pain to establish test-retest reliability. Path to Translation/Implementation: Our Smart Nerve Stimulator has strong paths towards translation towards commercialization as well as implementation into knee osteoarthritis research conducted at the CReATE Motion Center at the CMCVAMC and Atlanta VAMC. Our team will work with the University of Pennsylvania and CMCVAMC to protect our intellectual property and evaluate a path towards commercialization.

PROJECT SUMMARY/ABSTRACT Metabolic dysfunction-associated steatotic liver disease (MASLD) affects over 30% of the US population and has been identified as a leading cause of type II diabetes, hepatocellular carcinoma, and is now the most prevalent disease leading to liver transplantation. A defining feature of MASLD is the accumulation of unique triglyceride-rich organelles called lipid droplets (LDs). Understanding the fundamental mechanisms that regulate the hepatocellular storage, breakdown, and catabolism of LDs is essential to effectively prevent, reduce, and treat MASLD and is the focus of this proposal. Significant evidence, based on our work and others, implicates the selective targeting and breakdown of hepatic LDs by the autophagic machinery during a process called lipophagy. We recently identified a novel autophagic process termed “microlipophagy” (MiLi) by which lysosomes fuse, engulf, and degrade LDs directly. Our evidence indicates that MiLi is the predominant mechanism by which the hepatocyte catabolizes LDs. We also found that macropinocytosis (MP; “large cellular drinking”) plays a significant role in regulating hepatocellular lipid stores by forming large macropinosomes from the plasmalemma that traffic into the cell to mediate LD-lysosome fusion. We have demonstrated that important components of these essential cellular processes are large and small GTPases. These include the Ras-like Rab GTPases that control nearly all membrane-trafficking processes in the hepatocyte while the dynamin (Dyn2) family of large GTPase mechanoenzymes mediate membrane scission/fusion throughout the cell. Further, we have compelling evidence that this MiLi process occurs at the ER surface to catabolize nascent LDs as they form. Equally exciting to us is our finding that Mayo patients with MASLD possess mutant variants of these proteins that cause hepatocellular steatosis in culture. From these observations, the central hypothesis of this proposal predicts that together the MP and MiLi processes play a central role in hepatocellular lipid catabolism and are both supported and regulated by the synergistic actions of specific Rabs (Rab8a/10) and Dyn2 GTPases, which are altered and disrupted during steatosis. The strategy of this proposal utilizes state of the art hepatocellular imaging approaches, coupled with electron microscopy, molecular methods, and membrane biochemistry. This is correlated with data gleaned from patients, and 4 distinct and novel conditional knock out mouse models we have designed. Aim 1 will define the physiological contributions of MP to hepatocellular lipid stores and steatosis by testing how MP drives MiLi via a novel protein complex of Rab 8a/10, the large GTPase Dyn2, and a new endocytic adapter (SH3D19) we have identified. Aim 2 will define the mechanisms of a novel process we have observed that is focused on lysosomal targeting and catabolism of nascent LDs at the ER as they form that utilizes the actin cytoskeleton and ER-phagy/autophagy receptors. Completion of these studies will provide valuable insights into hepatocellular lipid metabolism, the underlying basis for hepatic steatosis, and potential novel strategies for therapeutic intervention in MASLD.

Ballard Public Art Committee

The Ballard Alliance, Ballard Public Art Committee, and Parks and Rec (the parties) aim to increase community ownership of Ballard Commons Park, the only open green space in the core of downtown Ballard. For years, the parties have collaborated on free park activities and have developed plans for a children's play area in the park which will break ground this year. For this grant proposal, the parties aim to conduct a community painted mural on a utility building in the NW corner of the park with a local artist during the Seattle One Day of Service. This project was originally proposed in 2020 but did not receive funding due to the pandemic. For this project, the artist will paint the outline of the mural on the utility building prior to the One Day of Service so that during the event, volunteers will fill in the mural in a paint by numbers approach. The mural, with existing park activities and future children's play area, will benefit the park and the community it serves.

The Ballard Public Art Committee will incorporate more public art on the streets of Ballard through the Utility Box Beautification Project. Located between 17th Avenue NW and 24th Avenue NW, there are six Seattle Department of Transportation utility boxes on NW Market Street that the committee will cover with vinyl wrap art. NCI - National Cancer Institute

ABSTRACT Biomarkers of exposure and biological effects are invaluable tools for studying the etiology of environmentally- induced cancers, identifying individuals and populations at risk, and developing and evaluating preventive interventions. This proposal will leverage the expertise and the teaching and capacity building experience of the Masonic Cancer Center (MCC), University of Minnesota faculty to provide an innovative, hands-on course to develop skills necessary for employing biomarkers of carcinogen exposure and biological effects in studies of cancer etiology and prevention. The course is targeted towards researchers-in-training (RITs) such as predoctoral and medical students and postdoctoral trainees from the US and from countries that represent high burden of exposure to environmental and lifestyle cancer risk factors. We will recruit 10 RITs each year, with each cohort consisting of four US-based participants and – leveraging our existing partnerships – six participants from India, Thailand, and African countries (two participants from each site). Specific Aims are (1) Provide a course that offers structured didactic content and hands-on experiences for developing skills in biomarker measurement and data analysis and interpretation. The course will include virtual modules focused on the fundamentals of chemical carcinogenesis and examples of biomarker development and applications, and a 5- week in-person laboratory training in state-of-the art biomarker measurement techniques. (2) Provide course participants with an understanding of clinical and translational biomarker applications to inform cancer prevention and control. Participants will learn and practice methodologies for designing, planning, and implementing human cross-sectional and intervention studies employing biomarkers. This will include modules on the ethics of human subject research as well as the logistics of project management, such as participant recruitment, retention, compliance with study procedures, and biological sample collection, processing, and shipment. (3) Provide course participants with skills and opportunities to establish global collaborative partnerships for conducting biomarker-based research in the area of cancer prevention and control. Each participant will partner with a participant from a different country to develop a joint research proposal employing biomarker assessments. The proposal will address a cancer risk factor relevant to both countries (e.g., air pollution, tobacco, diet) and will be designed to ensure that its findings will serve to inform each country’s cancer prevention and control efforts. Our long-term goal is to establish a community of highly skilled researchers who can engage in collaborative, translational biomarker research aimed to inform policies and practices to reduce the global burden of cancer.

This project explores the mechanisms by which cells respond and adapt to biochemical and physical cues from their environment. A particular focus is placed on mechanotransduction signaling in response to the material properties of a cell’s environment, leading to the nuclear translocation of important transcription regulatory proteins such as yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ). The project aims to quantify cell response to substrate nanotopography (i.e., nanoscale surface features) and to molecules that activate G protein-coupled receptors (GPCRs). It is hypothesized that cells integrate physical and chemical environmental cues through intracellular signaling events at the cell and nuclear surfaces, leading to distinct changes in YAP/TAZ localization and in cellular morphology. Throughout, this work utilizes a productive cycle of biophysical experiments and simulations. The K99 phase of this project examines cell and nuclear deformation on substrates with nanoscale topographical features, in the presence or absence of the GPCR agonist angiotensin II. Experiments will quantify the response of a human cell line (U2OS) to substrates with nanopillars. Mechanotransduction signaling will be quantified by immunofluorescence imaging of cytoskeletal activation, nuclear deformation, and YAP/TAZ localization in fixed cells, while biosensor-based fluorescence imaging will be used to monitor GPCR activation and downstream signaling events in live cells. These data will constrain a computational model that considers spatiotemporal dynamics of biochemical signaling within the cell coupled to curvature of the plasma membrane and deformation of the nucleus. Such simulations leverage recent advances in finite-element-based software packages used for spatial models of cell signaling and cell mechanics. This work will be conducted at the University of California, San Diego, supported by state-of-the-art experimental and computational resources and a strong team of scientific advisors including co-mentors Profs. Padmini Rangamani (primary) and Jin Zhang. This team will support training in biophysical experiments and computation. The candidate’s career goal is obtaining a tenure-track faculty position to lead research using both experimental and computational approaches to understand the biophysics of mechanotransduction. The associated R00 research will use human neutrophils as a model cell. Cell spreading and associated signaling events will be monitored in the presence or absence of the GPCR agonist CXCL8 on flat substrates of varying stiffness to assess how substrate deformability influences spreading efficacy. These results will be leveraged against predictions of a fluid-mechanics-based model of neutrophil motility coupled to a biochemical model of integrin-mediated signaling in neutrophils. Finally, deformation of the lobed and flexible neutrophil nucleus will be measured on nanopillar substrates and compared to that of rigid nuclei. Coupled mechanochemical simulations will be used to assess the implications of nuclear deformability for mechanosensitive signaling.

Black and Tan Project

On October 1st and 2nd of 2022, the Black and Tan Community Group will commemorate the 100 year anniversary of the opening of The Black and Tan Nightclub, at Langston Hughes Performing Arts Center. At the time, the first and only interracial nightclub in The Pacific Northwest. This historical event will consist of Two live performances will be held at the LHPAI followed by a reception. Our group is focused on bringing Arts, Culture and Theatre to the community free of charge! The event is free and open to the public.
