Abstract Hematopoiesis, the process of generating new immune cells (and other blood cells) in the bone marrow (BM), is precisely regulated by various niches, including adipocytes and their progenitors, to ensure immune competency and protection against pathogens. Adiponectin, the most abundant adipose-derived hormone, plays a crucial role in metabolic regulation; however, its function within the BM microenvironment remains largely unexplored. Obesity significantly reduces adiponectin levels, disrupting key physiological processes and increasing susceptibility to metabolic and infectious complications. While systemic adiponectin is recognized for its anti-inflammatory effects, its direct role in hematopoietic regulation and host defense against infections is not well understood. Our preliminary studies indicate that the BM adiponectin niche plays a direct role in B cell development beyond its well-established metabolic functions. Dysregulated BM adiponectin signaling compromises host defense, leading to increased susceptibility to infection-related complications. This study aims to characterize the BM adiponectin niche, identify its key cellular contributors, and determine how obesity alters adiponectin-driven cell-cell interactions within the bone marrow using adiponectin lineage-tracing models. Furthermore, we will employ receptor-deficient models to investigate how BM adiponectin signaling influences B cell hematopoiesis and impacts immune resilience. Through bone marrow transplantation experiments, we seek to assess whether adiponectin reconstitution can mitigate obesity-related disruptions in BM function. This research will provide critical insights into adipose-regulated mechanisms underlying metabolic adaptation and immune homeostasis, offering potential therapeutic strategies to address obesity- related complications. A deeper understanding of BM adiponectin’s role in metabolic and immune regulation may open new avenues for targeted interventions, ultimately improving health outcomes in obese individuals vulnerable to infection and metabolic dysfunction.
